How are calories calculated?

It seems that calories burnt while bike riding are independent from the net altitude gain/loss. The same rides in opposite direction, one with a +500 net gain and the other with a -500 ft net loss came out with pretty much the same number of calories (the downhill direction actually came out with 20+ cal). So I am wondering why net gain is not taking into account? If I remember my high school Physics, work is function of distance and difference of height. Right?
